Yanofsky conducted experiments whereby he induced mutations in the trpA gene of E. coli . All 16 mutant alleles produced inactive forms on the enzyme. He showed that the mutational sites in the gene map for trpA appeared in the same order as the corresponding altered amino acids in the TrpA polypeptide chain. (ii) Analyses of mutationally altered proteins showed that a single amino acid changes at one time in one region of the protein. This was predictive of a non-overlapping code. An overlapping code would predict that a single base change would alter as many as three amino acids at adjacent positions in the protein.
