Name of the unknown from the Laboratory Key: Providencia Part 2 Score ________ /10 2.1: The appearance of the growth on the original agar slant of my unknown, Providencia, was more of a thin layer of yellow slime on top of the slant. 2.2: Once I received my unknown I proceeded to use the Gram stain test. The Gram stain consisted of me determining whether my unknown was Gram positive and Gram negative, by examining the unknown through a microscope after the stain is complete. I received the result of my unknown to be a Gram negative microorganism because it gave characteristics of being rod shaped organisms that was a pink color through the stain. I then tested my unknown through the lactose fermentation using the MacConkey plate and also the tryptone broth. After twenty four hours for both tests, I checked my MacConkey plate and saw that the unknown had produced yellow colonies on the agar which would give me a negative result to the test; and after applying the Kovac’s reagent to the tryptone broth, a red film appeared on the top layer of the broth giving me a positive result for that test. Therefore, comparing the results of my tests to the dendogram, this led me to believe that my unknown was the bacterium Providencia .
