I. Materials Needed a. 5 test tubes b. 1 mL transfer plastic pipettes c. Spectrophotometer d. Water bath e. Kimwipe f. Parafilm g. Enzyme h. EDTA i. Catechol j. PTU k. Citric acid l. Distilled water m. Potato n. Blender II. Create a catecholase enzyme solution (prepared by lab instructor) a. A chilled, chopped, and peeled potato will be place into a chilled blender i. 500 mL of chilled, distilled water should be added ii. Blend the contents for three, ten second bursts b. Strain the solution through cheesecloth and a funnel into a container i. Immediately seal and keep on ice III. Turn the spectrophotometer on a. Should be on for 15 minutes before use b. Wavelength should be set at 540 nm IV. Label tubes 1-5 and add contents as indicated below. a. Add the contents EXCEPT for catechol b. Pipette the chelating agents after gently mixing i. There will likely be settling c. Invert and shake the tubes every 2 minutes while the chelating agent and enzyme
