Section 8 Microbiology Lab Microbial Growth of two soils, B. megaterium, and C. violaceum with Ultraviolet Light Introduction Microbial growth is the cell division of a bacteria into two daughter cells. The process of bacteria doing this is call binary fission. Ultraviolet light or more known as UV light consists a very short wavelength (175-350 nm) is responsible for sunburn. In the visible spectrum is right under blue light which ranges from 450 to 500 nm. UV light kills many microorganisms if directly effected. It first it causes a mutation in the DNA. Two adjacent thymines bond together causing an incorrect base pair when replicated. This can cause many problems creating mutations. Mutations in the DNA can lead to the cells dying. Organisms on open surfaces and in the air are only killed by UV light radiation because it can not pass through plastic and glass. UV light is usually used in places where contamination can occur easily and cause risks. Some hospital rooms contain UV light to kill off unwanted bacteria. (1) The experiment that we performed tested the sensitivity of different bacteria under intense UV light. The sensitivity of the bacteria against the UV light, in this experiment, we compared non-spore forming cells (chromobacterium violaceum), two types of soil (garden and crop), and bacillus megaterium endospores. Different bacteria can be killed in different ways depending on the characteristics of the organism. Chemicals, starvation, drying, and heat can all have a different effect on the microorganisms. wavelengths of UV light, can repair itself even when damaged. This is called light repair. After the samples are exposed to UV light, they must be placed in dark container. (1) In the experiment it was important to read and understand the safety precautions. UV light can be damaging to our bodies, especially to our eyes and skin. Wearing safety glasses as a precautionary measure is important as well as making sure not to directly look at the UV lamps.
