Staining Why? Staining is used to make it easier to view bacteria using bright field microscopy . Staining provides contrast. Identify morphology, size, and arrangement of bacteria samples. Different types: Simple stain Differential stain Gram stain, Acid-fast- will retain the dye when heating. Structural stain Endospore

Acid-Fast Staining: Ziehl-Neelsen Method Acid-fast microorganisms have a high mycolic acid content in their cell walls Fastness means to retain the dye when challenged by a decolorizer Procedure: Carbol fuschin dye (Ziehl’s Carbolfuchsin –pink) is used with Primary Dye Acid alcohol is the decolorizer Methylene blue is the secondary dye Cerise (positive) Blue (negative) 3

M. tuberculosis M. leprae Nocardia Acid Fast Genera Mycobacterium Species = Mycobacterium tuberculosis and Mycobacterium leprae Nocardia partially acid fast procedure. Actually requires use of less stringent decolorizer than acid alcohol to achieve the same results.
