Lok Cheng Chemotaxis Lab Report Lab 1 2/20/2018 Observing Caenorhabditis Elegans behavior through the chemotaxis assay using Water, Lemon Juice and Wilton’s Butter Introduction: Caenorhabditis Elegans or more commonly known as C.elegans is an eukaryotic nematode that lives in temperate soil and is a pseudocoelomate; an organism that lacks a respiratory and circulatory system. It has a highly developed chemosensory system that allows them to detect chemical signals. 10% of C.elegans is made up of neurons which are involved in detecting chemical signals. This allows them to detect chemical signals that are involved with food such as bacteria and toxic conditions, allowing them to be attracted to food and allowing them to avoid harmful situations. (Anderson, Gary L., et al.)The Biological Process that will be used in this experiment is a Chemotaxis assay. Chemotaxis Assay is an experimental technique which is used to measure the capabilities of chemotactic abilities of different organisms. In this experiment, we will utilize this technique by putting two different substances on each side of the agar plate and observing how the chemosensory system of C.elegans reacts to it, and where it migrates to. Our first observations when conducting the experiment were that the C.elegans were attracted to Wilton’s butter and not Water. In the first experiment, though there were some anomalies, according to Figure 1, we could see that in almost all trials, the nematodes were mostly attracted to Wilton’s Butter. On average, there was an average of 10.25 nematodes attracted to the A side

and only 7.25 nematodes on the B side. The standard deviation for the first experiment was 0.63 which is relatively low, making our results significant, allowing us to conclude that the nematodes were attracted to Wilton’s Butter. This is relevant as this allows us to use Water as a control, knowing that the nematodes won’t be neither attracted or repelled by it. In the next experiment, we will be using the same biological process to test the chemotactic capabilities of C.elegans using Lemon Juice, Wilton’s Butter and Water. I hypothesize that if lemon juice is tested against water in a chemotaxis assay using C.elegans, then the C.elegans will move towards the water as lemon juice inhibits the growth of bacteria which is a source of food for C.elegans and creates an unfavorable environment for C.elegans.
