Immunocytochemistry Worksheet Biology 355 This is a confocal microscope image of antibody staining. There are (obviously) three colors. In these photos, blue is DAPI stain (not antibody) that finds DNA. The red staining is because of an antibody to a transporter protein (red is often, but not always rhodamine fluorescing) and green is a FITC-fluorescing antibody to tubulin. 1) What can you conclude about these images: a. Cell movement? Nothing, one snapshot in time. b. Cell density? Nothing, one microscopic field, don’t know about others. c. Cell shape? Presuming the transporter is located peripherally (good guess) and that tubulin is distributed throughout cell (also good guess), our cells are elongated. d. Protein localization? We can see where the transporter and tubulin are distributed in the cell, in a general way. e. Phase of cell division? Since tubulin is organized to make up the mitotic spindle during mitosis, I would evaluate this as NOT mitosis, but interphase.
