Two FDA approved drugs, Crestor (statin) and Lisinopril, are prescribed to him to lower his risk of stroke, heart attack and other possible cardiac complications. Also, your uncle received a message from his friend that consumption of licorice tea could lead to dangerously high blood pressure. Based on your understanding of the regulation of electrolytes and water balance, explain to your uncle how these three agents (Crestor, Lisinopril and Licorice) work biochemically and what the side effects of the FDA approved drugs are. Use diagrams to illustrate your points. In 2003, Mirkin CA invented an immunoassay called Bio-Bargodes (BBC) Assay and he published his ideas and findings in a paper entitled “Nanoparticle-based bio—barcodes for the ultrasensitive detection of proteins” in Science (2003 Sep 26; 301(5641): 1884—6). What is BBC assay? How does this BBC assay work to provide such a high sensitivity for detecting disease markers? As a chief scientific officer in a clinical laboratory, you plan to use aptamers to enrich the BBC assay to probe a new disease marker X. Write a description with illustrations to show the working principles behind and the advantages of using this approach for your subordinates. Apart from assay sensitivity, what other parameters you need to consider for assay development? The next step you plan to do is to prepare a monoclonal aptamer against the marker X by SELEX (Systematic Evolution of Ligands by Exponential Enrichment).
