Microbiology Lab EXAM 2 Study Guide: 1) Quadrant streak plate method: a) Purpose: of streaking for isolation of colony is to produce isolated colonies of an organism on an agar plate. This is useful when you need to separate organisms in a mixed culture or when you need to study the colony morphology of an organism b) How does it work? Know how to perform it. Streak plate technique is used to grow bacteria on a growth media surface so that individual bacterial colonies are isolated and sampled. * Agar plate * inoculating loop *Bunsen burner *Bacteria for isolation Hold the loop flat against the plate and streak across surface. Reflame the loop before changing direction of streaking the second quadrant. 2) Staining in Microscopy a) What’s the purpose of staining in microscopy Increase the contrast between the specimen and the background Determine cell morphology and arrangement differentiate between different specie b) Preparing a heat-fixed bacterial smear: purpose; how to heat fixed is the first step. Purpose: kills the bacteria makes bacteria adhere to the slide coagulates cytoplasmic proteins to make them more visible. Preparing a bacteria smear. Place a drop of water into the wax circle that has been created on the slide. (clean slide) using an inoculating sterile loop.

The ability to access any university’s resources through Course Hero proved invaluable in my case. I was behind on Tulane coursework and actually used UCLA’s materials to help me move forward and get everything together on time.
