Case Study 5 – Part II Haley Norman 1. The APP gene codes for a particular protein. Why does a mutated APP gene lead to problems in the brain? What are the two other genes (and their resulting gene products— the proteins) that, when mutated, can lead to EOFAD? How does each protein play a role in brain function? The APP gene gives instructions for making amyloid precursor protein. This is found in tissues and organs within the brain and central nervous system. A mutated APP gene changes one of the amino acids in the amyloid precursor protein. More specifically, valine is replaced with isoleucine. This leads to an increased amount of amyloid beta peptide (longer and stickier form of peptide). When these proteins fragments are released from the cell, amyloid plaques form in the brain. These plaques plus the buildup of amyloid beta peptide lead to the death of neurons. Ultimately progressing the symptoms to Alzheimer’s Disease.
