Ex. No. 1 Date: 16/03/2018 1.1 CALIBRATION CURVE AND LAMBDA ( λ ) MAX 1.2 AIM To determine the lambda (λ) max and plot a reliable calibration curve for Paracetamol. INTRODUCTION In pharmaceutical analysis, a calibration curve is a general method for determining the concentration of a substance in an unknown sample by comparing the unknown to a set of standard samples of known concentration. In spectrophotometric analysis, a series of standard solutions of known concentrations are prepared and absorbance is measured using spectrophotometer instrument to determine the unknown concentration of sample. In this experiment, Paracetamol powder is used as analysis to determine the lambda (λ) max. Paracetamol also known as acetaminophen, is an analgesic (pain reliever) and antipyretic (fever reducer) medicine. It is commonly used for the relieve of headaches and other minor aches along with pains and is a major ingredients in numerous cold and flu remedies. In combination of opiod analgesics, Paracetamol can also be used to in the management of pain such as post-surgical pain and providing palliative care. Though paracetamol is used to treat inflammatory pain, it is not generally classified as an NSAID because it exhibits only weak anti-inflammatory activity (1). 1.4 APPARATUS AND MATERIALS
Paracetamol powder UV-Visible Spectrophotometer Beaker Weighing balance Measuring cylinders Volumetric flasks Distilled water Distilled water dropper bottle Water bath Dropper Spatula Cuvette Filter paper Lens cleansing tissue 1.5 PROCEDURE Preparation of stock solution 1. 0.1M of paracetamol powder was calculated and prepared for stock solution. 2. 1.51 gram of paracetamol was weighed by using electronic weighing balance. 3. The powder was mixed in a 100 ml volumetric flask and was filled with 90 ml of distilled water and shake well. 4. Next, the mixture was placed in a water bath to dissolve the powder until a clear and homogeneous solution attained. 5. The solution is then removed from the water bath and kept aside until it cools down and the stopper was removed in between to release the pressure in the volumetric flask. 6. Finally, 10 ml of distilled water was added up to the mark on the volumetric flask to make it 100 ml. This solution was the stock solution. Preparation of serial dilution 1. 9 volumetric flasks which are dry and clean were taken and labeled serially as SD 1 to SD 9.
