2a: Culturing and handling of microorganismsINTRODUCTIONAseptic technique is required to transfer a pure culture from one vessel to another.It is a technique which uses practice and procedures to avoid contamination frompathogen. Many Petri plates and tissue culture plates were used for growing pureculture of microorganisms are made of plastic and come pre-sterilized from themanufacturer. Aseptic technique involves avoiding any contact between the pureculture, sterile medium, and sterile surfaces of the growth vessel with contaminatingmicroorganisms. Healthcare workers like doctors, nurses and pharmacists use aseptictechnique in surgery rooms, clinics, outpatient care centres and other healthcaresettings.When wanting to accomplish aseptic technique, the work are must be cleansedwith a disinfectant to reduce the numbers of potential contaminants. Besides that, thetransfer instruments are to be sterilized. As an example, the transfer loop is sterilizedvia heating with a Bunsen burner pre and post transferring. The work should beaccomplished quickly and efficiently to minimize the time of which contamination ofculture or laboratory workers can occur.Steps for transferring a culture from one vessel to another:1.Flame the transfer loop.2.Open and briefly flame the mouths of the culture tubes.3.Culture growths are picked up with the sterile loop and is transferred to a freshmedium.4.Flame the mouth of the culture vessel and reseal them.5.Re-flame the inoculating loop.The same technique is used for inoculating Petri plates but the plate is notflamed and for transferring microorganisms from a culture vessel to a microscopeslide.OBJECTIVE1. To develop a thorough understanding and knowledge about aseptic techniqueand culture transfer procedures.2. To learn how to safely transfer cultures of microorganisms from one containerto another without contaminating the culture or exposing yourself to them.
