Abstract Throughout the experimental process, we performed a series of steps which consisted of our gram staining technique. When performing this technique, we basically gathered our materials and recorded our procedures step by step. Afterwards, we then proceeded staining the three different staining dyes with the two specific specimens we had. These two specimens were B. subtilis and E. coli K-12 (nutrient broth) and we manipulated methylene blue, safranin, and crystal violet staining dyes for the experiment. Upon staining, we observed the various shapes, cell morphologies, and sizes of each specimen. They all appeared to have had a distinctive color and image. Thus, we were able to determine which one was gram-negative and positive based on our results. Above all, this gram staining technique helped to be able to distinguish between gram-positive and gram-negative cells.
