Lab 2 Discussion
The purposes of this experiment are to identify different forms of culture media,
to practice several techniques like how aseptic technique of inoculating bacteria is done,
to prepare culture media and calculate its ingredients, and to know how to isolate
bacteria into pure culture using streak plate method. Results, for the autoclaved sample
show there was a growth of a colony in a white mucus irregular and wavy texture, and
non-autoclaved tubes had no growth. There was no growth of different cultures media
on the complex and synthetic medium. The MacConkey plate showed growth of
E. coli
with red pinkish dot of colonies and
P.aeruginosa
with light pink clear colony. While the
soduim Azide plate showed growth of
E.faecalis
with whitish doted colonies and
S.aureus
with clear colonies. The isolated bacteria into colonies to test the pure culture
technique showed no contamination and little to no single colony isolation, and the first
and second quadrant were not fully covered with colonies.
