Ch 19 Outline Recombinant DNA technology  is a set of techniques that locate, isolate, alter, and study DNA.  Sometimes it is  also referred to as genetic engineering or biotechnology. I.  Basic Molecular Techniques A.  Restriction Enzymes  (restriction endonucleases)  (Fig 19.2) make double-stranded cuts at specific sequences (palindromic sequences) isolated from bacteria Why do bacteria have them?  Why isn’t their own DNA digested? abbreviations from bacterial origin B.  Gel Electrophoresis—separates DNA fragments by size (Fig 19.3) transfers DNA fragments from gel to solid medium then a DNA sequence can be located by probing with a complementary sequence What is northern blotting?  western? D. Gene Cloning Produce identical copies (clones) of a gene How?  place gene into a host genome (bacteria) to be replicated Cloning vector: carries foreign gene into host cell What are the requirements of a cloning vector?   What is a plasmid vector?  How can you insert a foreign gene into it? (Fig 19.5, 19.7)
