Lecture 20: Eukaryotic Gene Regulation 2 1. What is an enhancer? What is its function? What are the special properties of enhancers? Activators whos binding site is in DNA about 80-160 base pairs upstream from the start site; distance from start site doesn’t affect effectiveness; regulates RNA polymerase transcription of genes. 2. What are the two modules/domains of sequence-specific transcription activators? Activation domain: communication with general transcription factors/ Pol II -Acidic (E/D rich) -proline rich -glutamine rich -poorly structured DNA-binding domain: -highly structured -recognition of specific DNA sequences 3. Describe the structure of 3 distinct DNA-binding domains and draw schematic structure. helix-turn-helix (HTH) found in homeodomain proteins and other HTH variant proteins (winged- helix proteins, Pou domain proteins, and paired domain proteins); DNA-binding motif found in a variety of globular transcription factors. Basic region/Helix-loop-helix/leucine zipper (bHLHZip) DNA binding and dimerization motifs found in different combinations in a variety of coiled-coil/fibrous transcription factors that include:basic/leucine zipper (bZip) Basic/Helix-loop-Helix (bHLH), and basic-helix-loop- helix-zipper (bHLHZip) proteins; The basic region adopts a a-helical structure only upon binding to DNA “induced fit” Zinc (Zn) fingers: DNA-binding motif found in many eukaryotic proteins.
