Jordyn Stevens Agarose Gel Electrophoresis Study Questions 1. What is the most commonly used gel for electrophoresis and where does it come from? Agarose is the most commonly used gel for electrophoresis. It is an uncharged polysaccharide purified from agar, which comes from the cell wall of seaweeds, marine red algae in the family of rhodophyceae. 2. In relation to its size in base pairs, how will a DNA fragment migrate through a gel? Explain. In relation to its size, the DNA fragment will migrate inversely proportional through a gel. Large molecules will migrate more slowly through the gel than smaller molecules. The phosphate backbone of DNA is negatively charged so the DNA fragments will migrate through the gel to the positive electrode. At the completion of electrophoresis, DNA is visualized by staining. 3. Do circular and linear forms of DNA migrate the same? Why or why not?
