ELN Example Day #3: (Module 3: Enumeration of Microbes and Cells) (September 17, 2018) Terms : CFU -colony forming unit Aliquot -a portion of a larger whole, especially a sample taken for chemical analysis or other treatment Stock Solution -A stock solution is a concentrated solution that will be diluted to some lower concentration for actual use. Stock solutions are used to save preparation time, conserve materials, reduce storage space, and improve the accuracy with which working lower concentration solutions are prepared. v/v and w/v -this means the percent by weight of solute in the total volume of solution. Normally used where the solute is a solid. For example, a 10% w/v sodium chloride solution would be prepared by dissolving 10 grams of sodium chloride in a solvent (again, say, water) then making up to a final solution volume of 100ml Experiment 1: Growth Curve and Experimental Design Introduction Create an experiment that tests dilution of bacteria using BHI, a diluent, and bacteria. Using this information to great a growth curve. Protocol 1. Take a 8-well strip for your row and label the edge with row number 2. In this experiment, my row chose to test different mouthwashes to see which kills the most bacteria. For a total of 250 µl, put 25 µl of mouthwash, 11.25 µl of bacteria, and 213.75 µl of BHI into the wells 1-6.
