ELN Day #2: (Growing and visualizing microbes, pipetting) (September 10, 2018) Definitions Autoclave -A pressure chamber used to carry out industrial processes requiring elevated temperature and pressure different to ambient air pressure; used to perform sterilization Differential Media - Contains reagents (usually dyes) that react with pH changes or other metabolic consequences of growth to produce reactions that are indicative of particular species. Colonies will have species-specific characteristics, which are easily recognized. Examples include blood agar plates, mannitol salt agar and MacConkey agar. Isolated Colony - a colony which grows separate, without touching another colony on a solid medium Subculture - new microbiological culture made by transferring some cells from a previous culture to fresh growth medium Spectrophotometer - The spectrophotometer is an instrument which measures the amount of light of a specificed wavelength which passes through a medium. According to Beer's law, the amount of light absorbed by a medium is proportional to the concentration of the absorbing material or solute present. ISOLATING MICROBES Introduction To isolate bacteria using a streak technique on an agar plate. Protocol 1. Get a TSA agar plate and write on the media side, your initials, seat number, media type, date, and the test microorganisms. 2. Receive and open up the environmental plate with what you tested from last lab. 3. Using the Bunsen burner, get the inoculating loop and put it over the flame until the loop becomes red-hot.

ELN 4. Using the inoculating loop, take part of the sample from last lab that was incubated and transfer it to your new plate and streak it over the agar plate in one quadrant.
