DNA from wheat germ was digested with micrococcal nuclease and the resulting fragments were separated by agarose electrophoresis on a 1.5% gel. Lanes 2, 3, 4 and 5 contain samples of DNA that were digested with 20 uL of nuclease. The length of the incubation time of the enzyme with the samples varied. Lane 1 contains a 100 bp ladder. 1. Referring to Figure 1: Thoroughly explain the dark smear in the upper portion of lane three. 1 Nucleosome + Linker ~165 bp 3 Nucleosomes + Linker ~530 bp

Nucleosome Structure Data Analysis Jordyn Stevens The dark smear in the upper portion of lane three indicates that this lane contains the largest amount of undigested enzyme in the well. The smear also shows that this lane contains many long DNA fragments and histones of various lengths that cannot be resolved. 2. Referring to Figure 1: Thoroughly explain the dark band in the well of lane three. The dark band in the well of lane three indicates sample DNA and histones that were not digested by the nuclease. The dark band shows that the sample DNA and histones are too large to enter the gel. The darkness of the well may also indicate that it was overloaded, meaning sample DNA was put into the well and all of it cannot be pulled through the gel by the current. 3. Referring to Figure 1: On the gel , label a band that contains DNA fragments that were wrapped around one nucleosome plus the linker DNA [units should be in base pairs]. Also, label a band that contains DNA fragments that were wrapped around three nucleosomes plus the linker DNA. The third band up from the bottom represents the DNA associated with three nucleosomes.
