Minyak atsiri kulit buah jeruk Bali diekstraksi menggunakan metode destilasi uap dan komposisi senyawanya diidentifikasi menggunakan KG-MS ( Kromatografi Gas Spektrometer Massa). Uji aktivitas antibakteri dilakukan dengan menggunakan metode difusi agar terhadap bakteri Escherichia coli ( E.coli ) dan Staphylococcus aureus ( S.aureus ). Minyak atsiri yang diperoleh dari ekstraksi kulit buah jeruk Bali berwarna bening, memiliki bau khas seperti jeruk dengan rendemen 0,14 %. Hasil uji aktivitas menunjukkan minyak atsiri kulit buah Jeruk Bali dapat menghambat pertumbuhan bakteri E.coli dan S.aureus . Pada konsentrasi 50, 75, dan 100 ppm minyak atsiri tersebut memberikan daya hambat yang kuat terhadap E.coli dengan diameter hambat berturut-turut, 11, 14, dan 17mm. Namun aktivitas antibakterinya terhadap S.aureus pada 50 ppm tergolong sedang dengan daya hambat 9 mm, dan aktivitas yang kuat ditunjukkan pada konsentrasi 75 dan 100 ppm dengan daya hambat berturut-turut 11 mm dan 14 mm. Hasil analisis spektra KG-MS menunjukkan minyak atsiri kulit buah Jeruk Bali mengandung lima senyawa yang teridentifikasi sebagai senyawa α -pinen, mirsen, limonen, germak ren dan β -asaron. Kata kunci: Jeruk Bali, minyak atsiri, Staphylococcus aureus dan Escherichia coli ABSTRACT This study aimed to determine antibacterial activity of Citrus maxima peel essential oil and to analyse its chemical constituents. The essential oil was extracted using steam distillation method and its chemical constituent was identified utilizing Gas Chromatography-Mass Spectrometry (GC-MS) technique. Antibacterial activity testing against Escherichia coli ( E.coli ) and Staphylococcus aureus ( S.aureus ) bacteria was done using diffusion agar method. Steam distillation of fresh Citrus maxima peel gave 0.14 % colorless oil with specific aroma of citrus. The antibacterial activity testing result showed that the essential oil inhibited the growth of both E.coli and S.aureus bacteria. The essential oil at 50 ppm; 75 ppm; and 100 ppm strongly inhibited the growth of E.coli with inhibition diameter of 11mm; 14 mm; and 17 mm and S.aureus by 9 mm;11 mm and 14 mm respectively. GC- MS spectra showed 5 peaks which identified as α -pinene, mircene, limonen, germacren and β - asaron. Keywords : Citrus maxima , essential oil, Staphylococu s aureus, Escherichia coli.
