Colombo No. 1 Yogyakarta e-mail: [email protected] Abstrak Penelitian ini bertujuan untuk menentukan kondisi optimum enzim tripsin dan mengetahui pengaruh penambahan ion logam Ag + (dalam bentuk senyawa AgNO 3 ), ion logam Cu 2+ (dalam bentuk senyawa CuCl 2 ), ion logam K + (dalam bentuk senyawa KHPO 4 ), dan ion logam Zn 2+ (dalam bentuk senyawa ZnSO 4 ) dalam berbagai variasi konsentrasi terhadap aktivitas enzim tripsin dengan substrat kasein. Aktivitas enzim tripsin dengan substrat kasein ditentukan dengan metode Anson pada kondisi optimum. Analisis data secara deskriptif kualitatif. Hasil yang diperoleh menunjukkan aktivitas enzim tripsin dengan substrat kasein 10 mg/mL pada kondisi optimum pada pH 8,37°C, waktu inkubasi 20 menit dengan penambahan ion logam (Ag + , Cu 2+ , Zn 2+ , K + ) pada berbagai variasi konsentrasi. Berdasarkan hasil penelitian terbukti secara empiris adanya kecenderungan ion logam Ag + dan Cu 2+ bersifat inhibitor dan ion logam Zn 2+ dan K + bersifat aktivator terhadap aktivitas enzim tripsin pada substrat kasein. Kata kunci : ion logam, kasein, tripsin, aktivator, inhibitor Abstract This study was aimed at determining the optimal conditions of the trypsine enzyme and the effect of metal ion Ag + (in the form of AgNO 3 ), metal ion Cu 2+ (in the form of CuCl 2 ), K + metal ions (in the form of KHPO 4 ), and ion Metal Zn 2+ (in the form of ZnSO 4 ) in various concentrations towards trypsin enzyme activity using casein substrate. The activity of trypsin enzyme was determined by Anson method in optimum conditions. The data were analyzed using qualitative descriptive. The results show that the activity of trypsin enzyme with substrate casein 10mg/mL in pH 8.37°C, incubation time of 20 minutes with the additional of ion (Ag + , Cu 2+ , Zn 2+ , K + ) at various concentrations. Based on the research results, it was empirically proven that they were tendency of Ag + dan Cu 2+ to act as inhibitors and metal ions Zn 2+ and K + to act as activators to trypsine enzyme activity at casein substrat. Enzim dapat dihasilkan oleh hewan, tumbuhan dan mikroorganisme. Secara katalitik, enzim menjalankan fungsinya dalam berbagai reaksi seperti hidrolisis, oksidasi, reduksi, isomerasi, adisi, transfer gugus, dan kadang-kadang pemutusan rantai karbon (Sumardjo, 2006). Enzim telah banyak digunakan dalam berbagai proses kimiawi, baik dalam bidang industri maupun dalam bidang bioteknologi. Seiring dengan peningkatan penggunaan enzim, berbagai eksplorasi penelitian tentang enzim telah banyak dilakukan (Falch, 1991).
