1 Zainab Makki NFS 5140-005,006 November 4, 2013 Methylene Blue Reduction Test in Milk for Microbial Growth Experiment 8 Abstract: Methylene Blue reduction test is an indirect measurement of oxygen. In this case, bacteria are responsible for the oxygen consumption in milk. Overall, the greater the number of bacteria present in the milk, the quicker oxygen will be consumed. As the oxygen is consumed, the color disappears. Colder milk, types of organisms, milk concentration, and incubation temperature will affect the reduction time. As the results show, according to this experiment, the color did reduce over time. After eight hours, the bacteria in the milk reduced by the methylene blue dye to a milky light blue. If the tubes were left for a longer period of time and hopefully at a higher incubation temperature the methylene blue dye would most likely be reduced to almost white. Introduction: Methylene Blue is a dye and it is an aromatic chemical compound having the molecular formula C 16 H 18 N 3 SCl. It is a dark green powder at room temperature and when it is dissolved in water, it forms a blue solution. The methylene blue reduction test is based on milk color when mixed with a dye.
