1 Zainab Makki NFS 5140-005,006 November 18, 2013 Phenol-Sulphuric Acid Assay (Total Carbohydrate Analysis) Experiment10 Abstract : Carbohydrates are hydrated forms of carbon and only contain the elements carbon, hydrogen and oxygen. They come in the form of (CH 2 O) n. . Carbohydrates are used in numerous ways, such as, metabolism of plants and animals, sources of sweetness, stabilizers or thickeners, fat replacers and as chromatographic material in pharmaceutical industries. The Phenol-Sulfuric Acid method was used in this experiment to determine the total concentration of carbohydrates present in a sample. A clear aqueous solution of carbohydrates to be analyzed are added to microtiter plate, then phenol and sulfuric acid are added. The absorbance is proportional to the carbohydrate concentration initially in the sample. The absorbance was largest at 100μg/μL and least at 0μg/μL. As for the unknown, it had an absorbance almost identical to that of 40μg/μL. Introduction : Carbohydrates are the most widely distributed and abundant organic compounds. They are the central role in metabolism of plants (photosynthesis) and animals (glycolysis). Carbohydrates provide 70-80 percent of calories in the diet worldwide. Carbohydrates contain only the elements carbon, hydrogen, and oxygen with the basic formula (CH 2 O) n . Carbohydrates have many purposes. They can be a source for sweetness, such as carbohydrates sucrose and fructose. They can be used as stabilizers or thickeners; pectin, gums, and modified starch.
