OBJECTIVE 1. To measure the effect of enzyme concentration on the rate of reaction. 2. To measure the effect of substrate concentration on the rate of reaction. 3. To measure the effect of pH on enzyme activity. MATERIALS Distilled water, amylase, starch, pH APPARATUS Spectrophotometer, cuvettes, dropper, marking pen, ruler, and parafilm squares. PROCEDURE a) Calibration and setting for spectrophotometer 1. The cuvette was measured up to 2 cm from the bottom of the cuvette and marked using marking pen. 2. Distilled water was added to the 2 cm mark for each cuvettes. 3. 10 drops of starch was added using dropper. 4. The cuvette was covered with parafilm squares and the solution was mixed up by invert the cuvettes ups and down. 5. The absorbance was read using spectrophotometer at wavelength 410 nm. 6. Absorbance was recorded.
