Primary Literature Article: Loss of mdig expression enhances DNA and histone methylation and metastasis of aggressive breast cancer Introduction/Background What is the mdig gene, and what type of samples have shown its expression? o The mdig gene (mineral-dust induced gene) is a lung cancer associated oncogene. o It was discovered in alveolar macrophages from coal miners exposed to mineral dust Which cancers has mdig been shown to expressed in? o Primarily expressed in lung and breast cancers What role has mdig been suggested to have in cells, and is there any information on its possible signaling pathway/mechanism? o Cell proliferation, neoplasias, pulmonary inflammation, and immune regulation What is the difference between a demethylase and a methyltransferase (DNMT)? o Demethylases remove methyl groups from molecules, and methyltransferases catalyze a transfer of a methyl group from the donor to their substrates What are the main findings that the authors are claiming in their introduction? o They are claiming that mdig regulates cell growth, and migration/invasion through DNA and histome methylation. o Mdigs oncogenic role may be dependent on the stage of cancer Earlier stages of development – mdig is oncogenic and promotes growth of tumor cells Later stages – may be inhibitory to metastasis of cancer cells Experimental Design Make sure that you understand they type of experiments that were used in this study. Be able to explain how each experiment works and why it would be useful for this study. o Cell Culture o RT-PCR i. This is a technique used to detect RNA expression ii. In this experiment, it could show the expression of mdig in each cell line, as seen in figure 1.
