Oct. 3 Citric acid cycle is on anaerobic pathway - Oxaloacetate = most important intermediate Anaplerotic pathways of tca OAA must be replenished for the cycle to continue - Anaplerotic – used to produce intermediate of the tca cycle Incomplete reductive tca pathway - To produce oxaloacetate, succinyl- coa and a-ketoglutarate - -some fermenting bacteria lacks a- ketoglutarate dehydrogenase no conversion of a ketoglutarate to succinyl coa - observed also in some bacteria (during anaerobic respiration) a-ketoglutarate dehydrogenase level is low under anaerobic respiration reductive citric acid (arnon-buchanan) cycle as it functions in green sulfur bacteria - produces oxaloacetate directly from citrate glyoxylate cycle modification oof the tca cycle being used to produce 4carbondicarboxylicacids from 2 carbon acetate units - key enzymes isocitrate lyase malate synthase - acetate increases activity of isocitrate lyase - glucose represses the key enzymes - acetyl coa from fatty acids, pyruvate and acetate can be used for gluconeogenesis isocitrate + acetate -> succinate + malate formation of PEP - essential for growth on lactate, [yruvate, acetate, succinate, malate and amino aicds 1. PEP from oxaloacetate PEP carboxykinase 2. PEP from pyruvate PEP synthetase -needs a mediator; formation of PEP from pyruvate formation of pyruvate from malate - Essential for growth on succinate or malate *Malic enzyme L-malate + NAD + <-> pyruvate + NADH + CO 2 (reduction nad to nadh and decarboxylation bc remove 1 carbon and becomes co2) Linking them all togethe The hub is the PDC (pyrate dehydrogenase complex) coverts pyruvate to acetyl coa for it to go to tca cycle. If not, pyruvate can be converted into other amino acids. Oxidative respiration: pdc is very important.
