132 PRODUKSI PROTEIN SEL TUNGGAL DARI KULTUR Rhizopus oryzae DENGAN MEDIUM LIMBAH CAIR TAHU SINGLE-CELL PROTEIN PRODUCTION THROUGH Rhizopus oryzae CULTURE WITH THE MEDIUM OF TOFU WASTEWATER Leny Maryana 1* , Syariful Anam 1 , Arsa Wahyu Nugrahani 1 1 Jurusan Farmasi, Fakultas MIPA, Universitas Tadulako, Palu, Indonesia Received 20 Agustus 2016, Accepted 28 September 2016 ABSTRAK Limbah cair tahu merupakan limbah hasil proses produksi pengolahan tahu yang dapat menurunkan kualitas lingkungan apabila tidak ditangani dengan tepat. Dampak lain yang dapat ditimbulakan adalah potensi terserangnya penyakit seperti diare, penyakit kulit, dan penyakit lainnya bagi masyarakat yang memanfaatkan air sungai yang telah tercemar limbah cair tahu ini. Akan tetapi, adanya kandungan karbohidrat dan protein yang masih tinggi pada limbah ini memungkinkan untuk dimanfaatkan lebih lanjut sebagai substrat pertumbuhan mikroba dalam menghasilkan Protein Sel Tunggal. Tujuan penelitian ini untuk mengetahui waktu inkubasi optimum dan kadar protein tertinggi Rhizopus oryzae pada medium limbah cair tahu berdasarkan perhitungan persen kadar protein dan analisis berat kering sel. Serta mengetahui pH optimum pertumbuhannya dengan variasi pH pertumbuhannya 4, 5, dan 6. Perhitungan dilakukan sesuai dengan waktu fermentasi yakni 24, 48, dan 72 jam. Kadar protein total dihitung dengan menggunakan metode Kjeldahl. Hasil penelitian menunjukkan kadar protein tertinggi terdapat pada masa inkubasi ke-48 jam dengan persen rata-rata protein yakni 0,47%, 0,47%, 0,46% dan massa sel dengan rata-rata nilai 0,77%, 0,84% dan 0,91%. Serta pH optimum pertumbuhan Rhizopus oryzae pada medium limbah cair tahu adalah pH 5.Dengan demikian dapat disimpulkan bahwa limbah cair tahu potensial digunakan sebagai substrat pertumbuhan Rhizopus oryzae. ABSTRACT Tofu wastewater is the origination of tofu’s production process that can degrade the quality of the environment if not handled properly. Another impact of this waste is the potential attack that could evoke the diseases such as diarrhea, skin diseases, and other illnesses for people who use the stream flow that has been polluted by this wastewater. However, the high-carbohydrate and protein of this waste may allow it to be used more as a microbial growth substrate to produce a Single-cell Protein. The purpose of this study is to determine the optimum incubation time and the highest protein content of Rhizopus oryzae in the medium of tofu wastewater based on the calculation of protein content percentage and the analysis of cell dry mass. And to know the optimum pH growth with the variation of pH growth which is about 4, 5, and 6. The calculation is performed in accordance with the fermentation time at 24, 48, and 72 hours. Total protein content calculated by the Kjeldahl method. The results showed the highest protein content obtained in the incubation period to 48 hours with an average percent of protein about 0.47%, 0.47%, 0.46% and a mass of cell with an average value of 0.77%, 0, 84% and 0.91%.
