UV light is capable of terminating most bacteria. O Used for sterilizing O Longer UV exposure should result in less growth (more damage/ death of cells) Different DNA repair mechanisms Photolyase Nucleotide Excision . Oxygen requirement o Thioglycollate medium (broth) 1. Aerobes= oxygen use (top) . Anaerobes= no oxygen (bottom) 3. Microaerophile= some oxygen use (center) 4. Facultative/Aerotolerant= growth with/without oxygen (throughout tube) . Temperature 1. Psychrophiles= -5C and 20C (cold ocean waters) 2. Mesophiles= 20C- 50C (majority, harmful pathogens) 3. Thermophiles= 50C- 80C (compost pile) 4. Hyperthermophiles= +80C (volcanoes) o Enzymatic activity decreases in lower temperature O Activity increase in higher temperature o Temperature also affect cellular membranes, transport, ribosomal activities of the cell.
