Crime Scene DNA Fingerprinting Emily Srisouvanh Introduction Within this lab experiment, the goal is to be able to determine 4 suspects of a crime scene utilizing multiple DNA samples to receive a match. In order for us to find our matches, we would have to use the polymerase chain reaction (PCR) and agarose electrophoresis, so that the results can be presented physically. The role of the electrophoresis can be a beneficial technique method used to find a DNA match through a fingerprinting process that is also known as “genetic fingerprinting”. We will try our best to discover the suspect of the hypothetical crime that was initiated. Methods Day 1 Gather 5 PCR tubes and label them CS, A, B, C, and D Add 10µl of MasterMix and 10µl of DNA (corresponding letters to the labeled tubes) Place them in the thermocycler (settings will provided in a picture under data) Day 2 Make a 1x TAE solution (10ml stock solution and 490ml of distilled water) Measure out 0.3g Agarose Combine the 0.3g Agarose with 30ml of your 1x TAE solution in an Erlenmeyer flask
