The sarcoplasmic reticulum acquires Ca 2+ via an active transport process driven by an ATPase pump. You have reconstituted the Ca 2+ -ATPase from the sarcoplasmic reticulum into pure phospholipid vesicles. Answer the following questions regarding the properties of these reconstituted vesicles: a. (5 pts) You find that the ATPase active site faces the outer surface of the vesicles. On which surface of the vesicle (in the vesicle lumen or on the outside of the vesicle) will you introduce Ca 2+ in order to observe ATP-dependent active transport? Explain your answer with a diagram. b. (5 pts) You find that the solubilized enzyme in a solution containing detergent is sensitive to vanadate and ADP. Is Ca 2+ transport in the reconstituted vesicle sensitive to vanadate? Explain. c. (5 pts) Using different conditions for the reconstitution of the Ca 2+ ATPase into vesicles, you find that the ATPase topology is changed such that ATPase activity is observed only when detergent is added to the vesicle preparation. Explain this behavior. Use a diagram to illustrate your answer.
