Assume that the critical concentration of an actin filament is 0.2 μ Μ , and is 0.1 μ Μ and 0.8 μ Μ for the plus and minus ends of the filament, respectively. (a). (6 pts) If actin is assembled to steady state in the presence of a protein that tightly caps the minus end of the filament, how much unassembled actin will there be at steady state? (b). (6 pts) A 10 μ Μ solution of actin is allowed to assemble to steady state, and is then diluted 100 fold. The level of assembled actin is monitored over time (solid line) beginning at the time of dilution. In a second reaction (dashed line), an actin- binding protein of an unknown activity is added at the time that the sample is diluted. Based on the affect on the disassembly kinetics, what activity might this protein have? Explain.
