Introduction to Media and Environmental Plates Mary G. Miller, Ed.D. Your Objectives this exercise: 1. Define media and the 3 forms associated. 2. Differentiate between active and dormant microbes. 3. Understand how majority of bacteria and fungi reproduce. 4. Identify common colony formations. Expand your understanding of the Microbial World Media is the plural form of medium. Media are forms of substrates, “food” for microbes, that are used to facilitate microbial growth out of the natural environment. In this course we will study a few fungi but will focus our study on bacteria. Because most bacteria are fastidious , only 10% of bacteria can be cultured outside of their natural environment. There are 3 forms of media. All three will be presented to you during your lab experience. Liquid medium is used to generate a “broth” of bacteria, also known as a broth culture. Broth culture is easily used to make smears for staining (explained in your next lab meeting) and is used most often in determining the metabolic abilities of the microbe being tested, such as fermentation tests described later in the semester. Semi-solid media , also known as stab or deep media, can consist of the same components in a liquid medium but with the addition of a range of .5-.7% agar, which is a solidifying agent.
