Unformatted text preview: Answers to Bio 118 in-class recitation problems (Sets 2-3) Set #2 Only one of the two DNA strands of the consensus sequence for the -35 box and -10 box of E.coli promoters is shown below: -35 Box: 5-TTGACAT -10 Box: TATAAT- 3 a) By convention, when only one strand of the DNA of a gene is shown (as it is in the above promoter) it is always the template strand or the non-template strand (circle one). Explain the reasoning behind this convention. Answer: THE NON-TEMPLATE STRAND IS USED BECAUSE IT HAS THE SAME SEQUENCE AS THE MESSENGER RNA. b) Below is a short stretch of DNA that is transcribed from right to left by RNA polymerase. The +1 base pair is indicated. Using the sequences above for the -35 and -10 box, attach these promoter elements to this DNA, by writing in the sequence appropriately, so that the enzyme, RNA polymerase, will be oriented correctly and know which strand of DNA to transcribe. You can use marks to indicate the spacing between the know which strand of DNA to transcribe....
