General Biology Lab #4- Enzyme Activity PLEASE NOTE: These questions/answers are only being e-mailed out because of lack of time during lab to properly explain everything. DO NOT copy these explanations into your LAB REPORTS!!! Everything must be written in your own words! Putting It All Together (Set #2): (Pgs. 128-134 of Text) 1) I. The following graph depicts the trend in data that you should have gotten for the absorbance (y-axis) per tube versus time (x-axis) in both sections of Part II of the lab (addition of enzyme and substrate). Note that as the concentration of either substrate or enzyme decreased (from Tube #1-#6) the rate of the reaction (slope of graph) also decreased. Tube #1 in both scenarios had the highest concentration of enzyme or substrate, and therefore had the highest rate of reaction (or absorbance per time) because there was plenty of enzyme or substrate to interact with their counterpart. However, by Tube #6 the amount of either enzyme/substrate was very small in comparison and so the reaction rate (or absorbance per time) was fairly decreased. Therefore the less substrate/enzyme concentration available for a reaction, the slower the reaction rate or the less absorbance over time will take place. Tube 1 = Black (top) Tube 3 = Red (middle) Tube 6 = Blue (bottom) II. In order to find the rate of a reaction, use the rate equation on page 3 of your lab manual: ∆A = A f -A i = rate of reaction ∆ t t f -t i **Please note, if you are looking to find the rate of your reaction per ten minutes (or any other amount of specified time) you need to use the above equation to find your rate for your reaction per minutes of your own rxn. and then multiply that amount by a factor which will give you a total of 10 min. in your denominator . (i.e. if you gat a rxn rate of 4A/5 minutes you would need to multiply that rate by 2 to
