Midterm 2 Bacterial Genetics- conjugation, transformation, and transduction. Transduction: phage-lytic: generalized Lysogenic: specialized Complete media: everything needed to grow. Minimal media: wild type can make additional components and grow. Bacterial colonies each derived from a single cell. Suspension of bacterial cells-> suspension spread on Petri plate with agar gel-> incubate from 1 to 2 days and single cells (not visible to naked eye)-> visible colonies (each a clone of the corresponding single cell). Single colony: one bacterial cell in one region that copies to many cells. Prototroph: can live on minimal media. Auxotroph: can’t live on minimal media. Resistance: resistant to an antibiotic trp- trp A, B, C Replica plating-> store master plate-> some cells move on the velvet covered surface-> Put in a replica plate and incubate with minimal medium. Selection: only a specific type of mutant survives.
