Strategic Approach to Learning Instrumentation (SALI)    Strategy for Using Spectrophotometer Orientation-Curiosity What is it for and what does it do? Used to measure the rate of a biochemical reaction and  the concentration of DNA or protein in a solution, to  quantify a change in color, and to count the number of  cells in a solution. It shines a wavelength of light onto  biological samples and measures the amount of light that  passes through a sample. What components make up the  equipment and how does it work? A light source emits radiation at different wavelengths. A  tungsten lamp is used for light in the visible range and a  deuterium lamp is used for light in the UV range. A  monochromator consists of a prism or diffraction  gratings which selects light at a particular wavelength.  The spectrometer produces a light of a selected  wavelength. The cuvette is used to hold the sample. The  detector is a photoiodide that measures the intensity of  light transmitted and sends a voltage signal to a display  device. The photometer measures the light intensity. Caution-Safe Use of Equipment Danger The cuvette may contain a solution that is deleterious if  skin contact occurs.  Breakable Handle the cuvette properly since it is made of glass, thus  is very fragile. Electrical n/a Planning n/a Start-Up  Rotate the power knob to turn-on. Once turned on, let  the machine warm up for 10 minutes before use. Calibration
