Ashkan Sajedi MCIM 417 Dr. Wilson 11225055 September 18 th , 2019 Assignment #1 The 2 methods we use to begin to characterize how the new virus enters cells are: Method #1 – Our unknown virus can enter the host cell through penetration and uncoating at the cell membrane. This pathway is for non-enveloped viruses which begins with high affinity interactions between the virus and the host cell receptors. This in turn induces conformational changes in the virus protein structures which then activate a signalling cascade that destabilize the host cell plasma membrane leading to pore formation and virus entry/internalization. This method of virus entry is pH dependant and it occurs at a neutral pH. We use fluorescent imaging and the use of specific inhibitory drugs to test our hypothesis. The unknown viruses are fluorescently labelled and infected with a culture of cells, through which the fluorescent imaging will show us if the virus forms an endosome upon binding with the host cell. If there is an endosome shown that would mean that the virus is using an endolytic pathway to enter the cell. If
