Site-directed mutagenesis • Alters the DNA sequence of a gene in a directed way such that amino acids are changed in the protein product according to the experimenter’s needs. • Used to test “what if the protein sequence was different from the normal sequence?”

Protein expression in bacteria • Many proteins are not present in high enough quantities (in their normal cellular environment) for us to purify and study them. • By moving the gene coding for this protein into bacteria, we can produce large quantities of the protein to study (enough to crystallize, for example).
