Midterm Sample Exam Answers Note: These questions / answers are designed to give you an idea of the types of questions that will appear on the midterm. The actual content and subjects may differ, since subjects are being covered in a slightly different order from previous quarters. Therefore, some of the questions appearing on the sample exam have not been covered yet in fall, 2007. 1. a) Phase contrast microscopy will allow you to see and evaluate the living, unstained cells. b) Yes, since phase contrast light microscopy, with the proper lenses and set up, can resolve objects as small as 0.200 um. c) Separate RER from SER by equilibrium density gradient centrifugation; based on differences in buoyant density between the two membranes (with and without attached ribososmes). d) TEM of a section through the isolated fractions. 2. Experiment: - obtain pure population of undifferentiated ECs (PECAM-1 positive; EpohrinB2 negative) - place both cell types in culture together for 2 – 3 days.
