Lecture 19 Highlights Baldin et al - Cyclin D1 Describe how Baldin et al synchronize fibroblast cells. First, how do they stop the cell cycle? Then, how do they start it? Describe how Baldin et al measure the percentage of cells in S phase. (i.e., what is BrdU and how is it detected?) Be able to interpret a plot of percentage of BrdU positive cells over time. I.e., be able to draw conclusions about when cells begin to enter S phase and about what percentage of cells are in S phase by looking at data such as fig 2A. Understand the strategy of a pulse labeling experiment. Interpret data in Figure 2b versus figure 2C. (e.g, figure 2 b shows you that the steady state level of cyclin is low during what stage and high during what stages?. .... . and 2c shows you that the synthesis of cyclin begins at. ....... does either method allow you to distinquish between destruction of cyclin D1 versus stopping synthesis as a reason for the decrease in levels observed? Where is cyclin D 1 localized in the cell (Fig 4)?
