Chris Stanton Leah, Thursday Rm 41 Experiment 4 Procedure WEEK 1 1. You will be assigned 2 primer pairs out of a possible of 9 (labeled 1-9). You need 6 200 microliter PCR tubes with 6 labels, filling out the labels as shown: CSH61 Primer pair #x – initials CSH61 Primer pair #y – initials CSH54 Primer pair #x – initials CSH54 Primer pair #y – initials Exconjugate – Primer pair #x – initials Exconjugate – Primer pair #y – initials 2. Add 50 microliters of the correct PCR cocktail and primer pair to each of the six tubes. The mixtures contain a large amount of the 4 deoxyribonucleotides, Taq polymerase, and large amount of the two primers constituting each primer pair along with a buffer that allows the enzyme to work. 3. a. Using a toothpick, transfer a visible dollop of cells from a CSH61 colony to the appropriate tube labeled primer pair #x. Shake the toothpick around so that all the cells come off into the solution. b.
