Enzymes are Catalysts The binding of a substrate to an enzyme has no effect on the difference in free energy between the substrate and the product. This means that binding to the enzyme has no effect on the final equilibrium between substrate and product.

The effect of binding to an enzyme is to lower the activation energy required for transformation of the substrate to the transition state before it is subsequently converted to product. The reduction in activation energy is generally achieved by a much better fit of the transition state to the enzyme than either the substrate or the product.

Compounds which resemble most closely the transition state between substrate and product, rather than resembling either substrate or product, are generally bound much more tightly than either substrate or product, and typically act as powerful competitive inhibitors. These compounds are called transition state analogs.
