Marta Gebregziabher Analysis 10-Plasmid DNA and PCR 1. What causes the cell to lyse is the SDS because it has detergent which destroys the plasma membrane. The role of the alkalinaity is to put the DNA in “shock” so that it will separate. Both plasmid and chromosomal DNA are separated from double stand to single strand. 2. When alkalinity is brought back down to neutral pH, the proteins and large pieces of DNA precipitate. The large pieces of DNA precipitate because they have not anneled correctly. The plasmid DNA will not precipitate because it has anneled correctly and it is not weighed down by salts. 3. Ethanol will remove the hydration shell form the DNA. The DNA is surrounded by a shell of water. 4. Cations in the solution will facilitate the precipitation of the DNA because they will bind to the negative charged DNA and cause it to shrivel up. 5. Washed the pellet of precipitated DNA washed the left over salt. Also it will solublize the salt but not the DNA.
