Microbiology Major Unknown Fall 2009 Lab dynamics You will be isolating and identifying two unknown bacteria from a mixed culture using the morphological and biochemical methods with which you are already familiar. For any determinative work, it is absolutely necessary to obtain a pure culture. Therefore, the first priority is to isolate your two unknowns, before any biochemical tests are performed. A suggested time frame is below: First lab: Streak for isolation Second lab: Describe different colonies morphologically and subculture onto plates. Gram stain colonies picked for subculture. Third lab: Gram stain isolates to confirm purity and determine characteristics. Prepare working and reserve stocks. Inoculate FTM and motility. Design dichotomous key/flowchart (see lab manuals in class, Bergey’s, a n d W e b C T f o r i d e a s ) Fourth lab: Look at results and inoculate biochemical tests according to f l o w c h a r t .
