PLB 105 Lab Manual—Fall 2009 page 7-1 Laboratory 7 Phloem and Xylem Regeneration Experiment-part 3 Primary and secondary phloem tissue are topics of today's laboratory. Living and prepared material will be used to identify the cell types and the special features of sieve-tube members and sieve cells. In addition, you will complete the xylem regeneration experiment. A. Angiosperm Phloem 1. Cucurbita a. Cucurbita stem fresh tissue Make TS and LS of fresh Cucurbita stem tissue. Stain with aniline blue. Callose deposits surrounding sieve pores of sieve-tube members will stain blue. Observe the numerous sieve areas on both lateral and end walls. Look especially at the sieve plates and at the distribution of sieve pores and callose. Does callose also surround pores in the lateral sieve areas? Make a detailed drawing of one sieve plate. Also, sketch several sieve-tuber members and companion cells as they appear in this freshly stained material.
