BIOL 2040 20 TA Notes: Fungal Genetics 1. a) Score the asci in figure 6.4 and show your calculations for the gene-to-centromere distance. Here you must make the connection between the visual representation of the asci and the numbers you are working with. You must be able to distinguish (and count) the first and second segregation patterns, and the number of asci in which they occur. MI= 15, and MII = 7. The gene –to-centromere distance is 15.9 cM. b) Calculate the genetic distance between the ascospore colour gene and the centromere using the data you are provided with. Your TA provided you with “data”. There will be fewer second division asci (MII), because they are recombinant. For example: If MI= 73, and MII = 12, the gene –to-centromere distance is 7.06 cM. If MI= 40, and MII = 39, the gene –to-centromere distance is 24.68 cM. 2. Which property of the Neurospora crassa reproductive cycle allows it to be used for calculations of gene-to-centromere distance? Explain how this property makes it possible to do these calculations.
