Antineoplastic MOA -Affecting cell survival -Boosting the immune system in its efforts to combat the abnormal cells TYPES Alkylating Antimetabolites Antineoplastic Antibiotics Mitotic Inhibitors Hormones and Hormone Modulators Cancer Cell Specific Agents MOA React chemically with portions of the RNA, DNA, or other cellular proteins Have chemical structures like those of natural metabolites -Inhibit DNA production in cells that depend on certain natural metabolites to produce their DNA Not selective only for bacterial cells; toxic to human cells -Break up DNA links and others prevent DNA synthesis -Cytotoxic and interfere w/cellular DNA synthesis Drugs that kill cells as the process of mitosis begins -Inhibit DNA synthesis -Interfere w/ ability of a cell to divide; they block or alter DNA synthesis, thus causing cell death Used in cancers that are sensitive to estrogen stimulation -Some of them block or interfere w/ estrogen receptors sites to prevent growth of the cancer and cause cell death in some situations -used as antineoplastic are receptor-site specific or hormone specific to block the stimulation of growing cancer cells that are sensitive to the presence of that hormone Treat chronic myeloid leukemia (CML) and CD117-positive unresectable or metastatic malignant GI stromal tumors (GIST) -These drugs would not have the devastating effects on healthy cells in the body and would be more effective against particular cancer cells - there are three groups 1-protein tyrosine kinase inhibitors 2-Epidermal growth factor inhibitor 3- Proteasome inhibitor INDICATIONS -Lymphomas, leukemias, myelomas, some ovarian, testicular, and breast cancers and some pancreatic cancers -Tto of various leukemias and some GI and basal cell cancers -Squamous cell carcinomas, leukemias, Adenocarcinoma of the stomach and pancreas -Variety of tumors and leukemias - Breast Cancer postmeno women or in other women without ovarian function -Tto of Prostatic cancer that are sensitive to hormone manipulation -Leukemia -Gastrointestinal stromal tumor CONTRA INDIC -Pregnancy and Lactation -Any know allergy -Bone marrow suppression -Suppressed Renal or Hepatic function -Pregnancy and lactation -Individual w/ a known allergy to any of them -Bone marrow suppression, renal or hepatic dysfunction -Known GI ulcerations or ulcerative diseases Pregnancy and lactation -Individual w/ a known allergy to any of them -Bone marrow suppression, renal or hepatic dysfunction -Known GI ulcerations or ulcerative diseases Pregnancy and lactation -Individual w/ a known allergy to any of them -Bone marrow suppression, renal or hepatic dysfunction -Known GI ulcerations or ulcerative diseases -Pregnancy and lactation Hypercalcemia (Toremifene) -Known allergy - Renal or hepatic dysfunction -Pregnancy and lactation -Hepatic dysfunctio -Pt w/ risk for prolongated QT intervals (hypokalemia, Hypomagnesia or taking drugs that prolonging QT - History of Hypersensitivity.
