6 Aharon’s 3 rd Exam Review Packet (Lectures 20 – 24) Disclaimer: This packet is a GENERAL idea of what you need to know for the 3 rd exam. It does NOT have everything you need to know for the exam. GOOD LUCK . Also, for lectures 20-24, focus on the mechanisms discussed in class, putting greatest emphasis on the transition states/covalent intermediates Cell Respiration Overview Red Arrows = energy transfer Black Arrows = Carbon compound movement

Lecture 20 and 21 - Glycolysis Watch the following video, it does an awesome job explaining Glycolysis Video: http://www.youtube.com/watch?v= O5eMW4b29rg (step by step video) You should know: History of glycolysis i.e. what did Buchner brothers, Harden and Young, and Pasteur did How dialysis works in terms of glucose and its necessary enzymes and cofactors. Also, know where you would end up if you missing necessary enzymes, cofactors What a zymase and cozymase are Be able to recognize and know how to use NAD How to draw all structures and follow each carbon throughout the glycolytic pathway Every enzyme for each step What cofactors are necessary for each step (ATP, NAD + , Mg 2+ , etc.)

All intermediates between each step and the mechanisms of each step o What type of intermediate are created (eg. Schiff Imine base) Which steps are reversible or irreversible and why? Competitive inhibitors of certain substrates that bind tightly and represent transition state Understand overall equation of gylcolysis o Glucose + 2ADP + 2Pi +2NAD + ---> 2pyruvate + 2ATP + 2NADH + 2H + + 2H 2 O Know experiments of proofs talked about in lecture (eg. GAPDH mechanism with Iodoacetate Know 3 different routes of pyruvate breakdown and if oxygen is necessary for each pathway o Know Lactic Acid and Ethanol fermentation including all necessary enzymes and cofactors.
