Ligand binding of Proteins The binding of a ligand [L] by proteins [P] can be expressed in binding curves. 1) Label axes. 2) Draw an independent binding curve for K D(A) = 1.0x10 -6 . (curve A) 3) What is Ɵ for [L] = 1.0x10 -6 ? 4) What type of curve is it? 5) What is the equation for Ɵ? What does Ɵ mean ( in words)? 6) Draw a graph (B) with a higher K D than A (K D(B) = 1.5x10 -6 ). 7) Is the binding affinity of protein B higher or lower than for protein A? 8) Draw a graph (C) with a lower K D (K D(C) = 0.5x10 -6 ). 9) Is the binding affinity of protein C higher or lower than for proteins A and B? 10) What is the relationship between K D and the ligand binding affinity of a protein? 11) Some proteins bind oxygen. The concentration of oxygen is usually expressed in partial pressure. Label x-axis for oxygen binding proteins. 12) What is p 50 (in words)? And what is its relationship to K

Unformatted text preview: D ? 13) Do graphs A, B and C resemble the binding curve of Myoglobin or Hemoglobin? Why? 14) Draw cooperative binding curves (where N>n>1, and n a =n b =n c same Hill Coeffcient) for the same K D s as above, label curves with lower case letters respectively.( e.g.(K D(A) = K D(a) ) 15) What type of curve is it? 16) Write an equation for Ɵ(O 2 ) as a function of pO 2 . 17) What is the value of Ɵ at p 50 ? 18) Is the oxygen affinity of Hemoglobin in the lungs high or low? Why? 19) What about the oxygen affinity of Hemoglobin in tissues? 20) Why does hemoglobin have different affinities for O 2 in lungs and tissue? 21) If you could synthesize a drug that would enhance your O 2 release in the tissues what would its function be? 1 0.5 1.0 1.5 2.0 2.5 0.5 3.0 3.5 (μM)...

What students are saying

• As a current student on this bumpy collegiate pathway, I stumbled upon Course Hero, where I can find study resources for nearly all my courses, get online help from tutors 24/7, and even share my old projects, papers, and lecture notes with other students.

Kiran Temple University Fox School of Business ‘17, Course Hero Intern

• I cannot even describe how much Course Hero helped me this summer. It’s truly become something I can always rely on and help me. In the end, I was not only able to survive summer classes, but I was able to thrive thanks to Course Hero.

Dana University of Pennsylvania ‘17, Course Hero Intern

• The ability to access any university’s resources through Course Hero proved invaluable in my case. I was behind on Tulane coursework and actually used UCLA’s materials to help me move forward and get everything together on time.
